Comparison of Anti-HCV EIA and Immunoblotting

Abstract
|

|
The exact siginificance of antibodies to hepatitis C virus(HCV) in blood donors remains unknown. Confirmatory tests of anti-HCV reactive serum are used to refute a large proportion of false-positive results. Distinct HCV antigens are applied
seperately
on nitrocellulose strips on which the anti-HCV antibody recognition patterns are made visible Forty-nine blood donors reactive for anti-HCV were tested with a second-generation(2nd gen)anti-HCV ELISA and a immunoblot assay Thirteen of 43(30.2%)
donors
with positive 2nd -gen ELISA tests were confirmed as positive by immunoblot. The specificity for the detection of HCV infection had increased in blood donors when the immunoblot assay was used instead of the 2nd gen ELISA Howerver, nine of
43(20.9%) 2nd
gen ELISA positive donors were immunoblot indeterminate It is concluded that the detection of HCV-RNA by reverse transcriptase-polymerase chain reaction(RT-PCR) seems to have resolved difficuities with immunoblot indeterminate pattern and false
positive
results in the 2nd gen ELISA .
